Abstract

Background

The aim of this study was to investigate the effect of the antiobesity drug orlistat on the metabolism of the most common type 2 diabetes mellitus in rats. Rats (n = 40) were treated with either orlistat (5, 10, or 20 mg/kg) for a period of 1 week, and the animals were observed for changes in food intake, body weight, glucose tolerance tests (GTs), and body weight. All the parameters studied (body weight, glucose tolerance, fasting glucose, and after food intake) were within the acceptable limits, and all the data were considered in a way that was meaningful to the model. Furthermore, the relationship between weight changes and glucose levels in plasma was assessed. The results suggest that orlistat increases the lipoprotein (PL) levels, insulin sensitivity, and GLUT-1 secretion in rats, whereas orlistat does not alter this response in rats.

Methodology

The animal study was carried out at the University of Central Brazil University Hospital in São Paulo, Brazil, between January and March 2021. All the experiments were performed in accordance with the Guide for the Care and Use of Laboratory Animals and the Brazilian Law on the Federal Office for the Protection of Laboratory Animals. The animals were randomly assigned to two groups: the control group (n = 40) and the orlistat group (n = 40). The control group was administered a single oral capsule (0.6 mg/kg) of the antiobesity drug orlistat (5, 10, or 20 mg/kg) for a period of 1 week, and the animals were observed for changes in food intake, body weight, glucose tolerance tests (GTs), and body weight. Furthermore, the relationship between weight changes and GLUT-1 secretion in plasma was assessed.

Results

The weight changes were significantly higher in the orlistat group than in the control group at both the beginning and the end of the 1 week period. There was no significant difference in the amount of body weight changes between the orlistat group and the control group at either the beginning or the end of the 1 week period. In addition, the change in the amount of body weight in the orlistat group was significantly lower than that in the control group at both the beginning and the end of the 1 week period. In addition, there was a significant positive relationship between the amount of body weight change and the GLUT-1 secretion in plasma. These results indicate that orlistat increases the lipoprotein (PL) levels, insulin sensitivity, and GLUT-1 secretion in rats, whereas orlistat does not alter these responses in rats.

Conclusion

Orlistat, a lipoprotein-lowering agent, increases the lipoprotein (PL) levels, insulin sensitivity, and GLUT-1 secretion in rats. The effects of orlistat on the PLs levels and GLUT-1 secretion were evaluated in rats. The results suggest that orlistat increases the lipoprotein (PL) levels, insulin sensitivity, and GLUT-1 secretion in rats, whereas orlistat does not alter these responses in rats.
